I am thinking of putting together the following layout which I would like to add 1 or 2 Google Adsense units in:
I would like to know if this would be allowed?
And also, if anyone has any suggestions to other formats that would work within this type of grid? I’m wondering if there would be a better way to display an ad, especially if it’s responsive.

The answer is the same. You need to ensure that the ads are clearly distinguishable as ads, and are not likely to be clicked by mistake.

I am still a bit confused as I think the ad is distinguished among the other content, e.g. the yellow boxes.
Would you say the ad would be easily clicked by mistake?
What google usually mean by these sorts of things is those ad banners that look like a download button on a ‘download this file’ page placed where you’d be looking for a download button when the actual download button is further down and less flashy. That’s the kind of ‘distinguished from the content’ sort of thing they’re trying to avoid.
