Nofollow links do not contribute to SEO or search engine rankings, if that is what you mean.

However, if you start contributing regularly to an online community, folks tend to check out your signature and profile and might visit links that appear in those places if what you say holds some value.

Computers are not people. If you spell a word wrong, and it’s part of the code, they will usually ignore it. If you put ‘nofolow’, to a computer that means nothing at all.

It looks like both Bing and Yahoo also respect nofollow.

Any relevant link whether it is follow or no-follow carries value. Keep things relevant and balanced though.

In respect to your other question, there are really no secret hacks that you can do in your code to help things up.
As a quick advice, make sure to add rich snippets around.
